Genetic linkage map construction and QTL mapping of blade length and width in Saccharina japonica using SSR and SNP markers

MAS (marker assisted selection) is useful for cultivar development in the lower plant Saccharina japonica . In this study, 167 screened SSR markers and 125 individuals were used to construct a S.japonica genetic linkage map with 27 groups total Haldane length of 2069 cM. Constructed aligned assembled pseudo-chromosomes S. genome sequences (NCBI: MEHQ00000000) anchored 23 31 pseudo-chromosomes. A including 33 was constructed combined 3397 SNP markers. integrated QTL mapping, marker M1895 closely associated major blade width LOD threshold >5.0. Annotation indicated that SjPT , coded as high affinity phosphate transporter, mapped cDNA sequence 1422 bp, which 473 aa 8 transmembrane helics. qRT-PCR transcriptomic detection confirmed transcription consistent increases kelp width. Marker appears be particularly breeding.
